Indications:

Tivoxen (Tivozanib) is indicated for the treatment of:

• Advanced renal cell carcinoma (RCC) in adults who have received one or more prior systemic therapies.
  Tivozanib is especially beneficial for patients requiring a highly selective VEGFR-targeted therapy.

Pharmacology:

Tivozanib is a highly selective tyrosine kinase inhibitor (TKI) that targets:

• VEGFR-1, VEGFR-2, and VEGFR-3 (vascular endothelial growth factor receptors)
  These receptors are responsible for tumor angiogenesis.
  By blocking them, Tivozanib starves the tumor of blood supply, thereby slowing or stopping tumor growth.

Dosage & Administration:

• Recommended dose: 1.34 mg once daily for 21 days on treatment, followed by 7 days off, in a 28-day cycle.

• Taken orally with or without food.

• Capsules should be swallowed whole, not chewed or opened.

Drug Interactions:

• Caution with CYP3A4 inhibitors/inducers.

• Avoid use with medications that prolong QT interval if possible.

Contraindications:

• Hypersensitivity to Tivozanib or any capsule component.

• Not recommended in patients with severe hepatic impairment.

Side Effects:

• Common: Hypertension, fatigue, diarrhea, decreased appetite, hoarseness.

• Serious: Blood clots, cardiac events, proteinuria, liver toxicity.

Pregnancy & Lactation:

• Not recommended during pregnancy; may cause fetal harm.

• Avoid breastfeeding during treatment and for 1 month after the final dose.

Precautions & Warnings:

• Monitor blood pressure, liver function, and thyroid function regularly.
