Indications:

Talaparib (Talazoparib) is indicated for the treatment of:

• Germline BRCA-mutated, HER2-negative locally advanced or metastatic breast cancer, in adult patients.
  Genetic mutation must be confirmed using an FDA-approved test prior to treatment.

Pharmacology:

Talazoparib is a PARP (Poly ADP-ribose polymerase) inhibitor.
It works by blocking PARP enzymes involved in repairing damaged DNA in cancer cells—especially in tumors with BRCA1/BRCA2 mutations. This leads to the accumulation of DNA damage, ultimately causing cancer cell death, while sparing normal cells.

Dosage & Administration:

• Standard dose: 1 mg orally once daily, taken with or without food.

• Capsules must be swallowed whole—do not open, crush, or chew.

• Dosage may be adjusted based on kidney function or side effects.

Drug Interactions:

• Caution with P-glycoprotein inhibitors/inducers.

• Monitor for enhanced toxicities when used with myelosuppressive drugs.

Contraindications:

• Known hypersensitivity to Talazoparib or any capsule component.

Side Effects:

• Common: Fatigue, nausea, anemia, neutropenia, thrombocytopenia, headache, diarrhea.

• Serious: Myelodysplastic syndrome (MDS), acute myeloid leukemia (AML).

Pregnancy & Lactation:

• Contraindicated during pregnancy due to fetal harm risk.

• Women should avoid pregnancy during and for 7 months after treatment.

• Avoid breastfeeding during and for 1 month after the last dose.

Precautions & Warnings:

• Monitor blood counts (CBC) frequently during treatment.

• Consider dose reduction or interruption if severe hematologic toxicity occurs.

Overdose Effects:

• May cause severe hematologic toxicity.

• Supportive care and monitoring are essential.
