📌 Indications:

Ibrutix (Ibrutinib) is indicated for the treatment of:

• Mantle Cell Lymphoma (MCL) in patients who have received at least one prior therapy

• Chronic Lymphocytic Leukemia (CLL) / Small Lymphocytic Lymphoma (SLL)

• Waldenström’s Macroglobulinemia (WM)

• Marginal Zone Lymphoma (MZL) in patients requiring systemic therapy

• Chronic Graft-versus-Host Disease (cGVHD) after failure of systemic therapy

💊 Pharmacology:

Ibrutinib is an oral, selective, and irreversible inhibitor of Bruton's Tyrosine Kinase (BTK), a key enzyme in the B-cell receptor signaling pathway.
By blocking BTK activity, Ibrutix:

• Inhibits malignant B-cell proliferation

• Promotes apoptosis (programmed cell death)

• Reduces tumor growth and survival

This targeted approach provides sustained disease control in B-cell malignancies with manageable toxicity.

💉 Dosage & Administration:

• MCL & MZL: 560 mg orally once daily (4 × 140 mg capsules)

• CLL/SLL, WM, cGVHD: 420 mg orally once daily (3 × 140 mg capsules)

• Capsules should be swallowed whole with water, at the same time each day

• Can be taken with or without food

• Avoid grapefruit and Seville orange products

• Dose adjustments may be necessary for toxicity or drug interactions

⚠️ Precautions & Warnings:

• Bleeding risk – use caution with anticoagulants/antiplatelets

• Infections – monitor for bacterial, viral, and fungal infections

• Cardiac effects – atrial fibrillation, ventricular arrhythmias possible

• Tumor lysis syndrome – monitor at-risk patients

• Avoid strong CYP3A4 inhibitors/inducers

❌ Contraindications:

• Hypersensitivity to Ibrutinib or any capsule component
